This study was carried out to determine whether Juglandis Semen aqua-acupuncture (JSA) exerts the protective effect against Hg-induced liver cell injury. The cell injury was estimated by measuring lactate dehydrogenase (LDH) release, and the lipid peroxidation was estimated by measurimg malondialdehyde (MDA), a product of lipid peroxidation, in rabbit liver slices. When tissues were incubated with 0.5mM Hg for 10¢¦120min, LDH release and lipid peroxidation were increased as function of incubation time, and these effects were significantly prevented by addition of 0.1% JSA. Hg increased LDH release and lipid peroxidation in dose-dependent manner over the range of 0.1¢¦1mM concentrations, which was reduced by 0.1% JSA. When tissues were treated with 0.5mM Hg in the presence of 0.05¢¦ 1 % JSA, LDH release and lipid peroxidation induced by HG were prevented by JSA in a dose-dependent fasion; 0.5% and 1% JSA prevented almost completely effects of 0.5mM Hg.
These results indicate that JSA exerts the protective effect against Hg-induced liver cell injury, and this effect is attributed to antioxidant action. However, the precise mechanisms of antioxidant effect of JSA remain to be defined.
